The semi-skirt frame includes orientation features that distinguish it from general 96-well PCR plates. The notch and corner pattern must align with the carrier, while thin-wall low-profile wells provide close block contact for thermal transfer.

Plate Structure and Functional Design

The semi-skirt frame includes orientation features that distinguish it from general 96-well PCR plates. The notch and corner pattern must align with the carrier, while thin-wall low-profile wells provide close block contact for thermal transfer.

Align the notches and corner orientation before closing the instrument lid. Never force a misaligned plate into the carrier, as this may deform the wells or compromise optical contact.

The notch pattern and skirt geometry are designed around selected LightCycler®-type instruments. Confirm whether your system is LightCycler 480, LightCycler 96 or another model; block and carrier requirements are not interchangeable.

  • Confirm the number of wells, block height, skirt style, corner cut and notch position.
  • For qPCR, confirm whether clear or white wells and an optical seal are required.
  • For automation, validate gripper access, deck carrier, barcode area and plate rigidity.

Sealing, Thermal Cycling and Handling

  • Use an optical adhesive film, heat seal or cap solution compatible with the plate and detection method.
  • Apply uniform sealing pressure and confirm that every well rim is fully closed before cycling.
  • Centrifuge briefly when the protocol requires liquid collection at the well bottom, using a supported carrier.
  • Keep wells and sealing rims free from powder, oil and fingerprints.
  • Nominal well capacity is not the recommended reaction volume; preserve headspace and follow the assay protocol.
